About

PeroxiPlat is the first small molecule fluorescence microscopy imaging agent that can be used to target peroxisomes in mammalian (including human) cells. PeroxiPlat is a compound designed for use in fluorescence cell imaging, combining a platinum (IV) trimethyl by pyridine core, which provides the fluorescence, with a glutathione unit, which provides the specific localisation within cells unique to this molecule. This novel methodology enables high contrast and high-resolution imaging of peroxisomes using confocal microscopy. PeroxiPlat can be used as a fluorescent imaging agent in live or dead (fixed) cells, offering an ability to image cells without influencing natural behaviours. Advantages 1) Current gene technology for peroxisome imaging interferes with the normal function of the cells may not accurately reflect peroxisomes in native cells. 2) PeroxiPlat is taken up and accumulates in peroxisomes within minutes of application. Gene technologies take several hours to a day to generate fluorescent peroxisomes 3) PeroxiPlat can be applied to cells from native samples. Therefore PeroxiPlat offers unique opportunities in diagnostic applications cell samples who would not have previously been transfected Malfunctioning or absence of peroxisomes is associated with many diseases, e.g. Zellwegger Syndrome Spectrum (ZSS), and peroxisome proliferation is linked with cancer.
