INVENTION Based on the proposition that amyloid fibril formation is pathologically associated with many neurodegenerative diseases, this invention presents a fluorescent bioprobe for the monitoring of amyloid fibrillation process. The nonluminescent tetraphenylethylene derivative (BSPOTPE) is induced to emit intensely due to the restriction of intramolecular rotation, showing a novel phenomenon of aggregation-induced emission (AIE). The AIE-active BSPOTPE exhibits distinct fluorescent behaviours toward the native and fibrillar forms of insulin and thus enables the quantitative analysis and kinetic investigation of the fibrillation process. Another observation is that premixing BSPOTPE with insulin can extend the lag phase and decelerate the growth rate of insulin fibrillation, demonstrating the role of BSPOTPE as an effective retarder for amyloid fibrillation. MARKET OPPORTUNITY The newly invented water-soluble AIE-active polyene compounds could be used for the detection and labeling of biomacromolecules. They are potential candidates for sensor device in biologic area. * As an indicator, the reagent can be used to monitor the fibrillation process of proteins. * As a retardant, the reagent can serve as a stabilizer for long-term storage of insulin for treatment of diabetes.
